Interesting. As you haven't mentioned or alluded to it at all, it is unlikely to the point of presumed impossibility, but I was wondering if Harry might be born some type of intersex. The most likely to me being Persistent Müllerian duct syndrome (PMDS) where the male has all the female plumbing save the outer bits which were all male. The one anon (dubbed "Rob" for simplicity's sake) I heard the story of even had periods. Having all the plumbing and fitting any other criteria we don't know for sure about that the Slayer essence uses as guidelines, an intersex male could feasibly become the Slayer, without magic accidental sex changes beforehand anyway.

Though honestly I feel the Slayer essence isn't too picky: fit most of the other criteria and then fit one or both of these boxes: box1- identity as female box2- possess female plumbing.
