Makes sense, sticks to characters, lovable OC's... It's awesome!

As an avid Fanfiction reader, I have to compliment your flawless work in the development of the OCs. The development of each characters background and how it plays into their feeling and actions towards life is very thought out.
My favorite OC would have to be Ash. I love the ideal wall she's built herself and how she chooses to indulge in things such as alcohol and hunting to relieve stress, which is a very Dean-like thing to do.
Although, I would like to see more of her denial towards her feelings for Castiel in chapter under her POV so, as a reader, we get more insight to why she tries to deny them to Lilly, Mira, etc.
I also can't wait for Ash to meet Jimmy during the time Castiel is "exorcised". I think they should have a personal moment to further develop her relationship with Castiel.

Lastly, I do have a question.
Knowing Ash can hear an angels true voice, was there a reason behind that for further plot development or was that written in to start the attraction Ash has to Castiel?

It's an amazing story :)

VERY nice bit of work, in my opinion.
Not only have you created some interesting OCs, but you’ve also provided them with a highly engaging backstory, avoiding the easy route of making them biological sisters to explore three different approaches to hunting, ranging from Lily as the more intellectual- more efficient at the rituals than the fighting-, Mira with trust issues, and Ashley simply more automatically hostile (Particularly given the circumstances of their meeting; even if they quickly established what the other was, that’s certainly one way to be intruding on a hunter’s territory).
Subsequent developments as they continued to meet up were also well-handled, gradually introducing the idea of the Winchesters and the three girls starting to hunt together as they find themselves running into each other during jobs without it ever appearing too coincidental or forced, particularly as you explored such elements as Mira’s ‘late arrival’ on the hunting scene (Considering that the other two were a team already) and subsequently revealed how Lily got into the hunting scene.
A part of me would have liked to see more of the two groups collaborating before Dean’s deal came through- as well as give them additional opportunities to bond outside of the occasional chance encounter during a hunt, the sisters could have run into the Winchesters and Gordon while tracking Lenore’s vampires, helped Dean deal with Meg-in-Sam, or become caught up in the demon siege while attempting to rescue the Winchesters from prison-, but them sticking together after the news about the Seals is revealed certainly makes sense, as does your idea to make Ashley able to hear angels as a means of giving her a particular connection to Castiel, to say nothing of her inner conflict over her reaction to his presence, to the point where she actually chooses the angels over the hunters at one particular point.
Still, all in all, you’ve made some interesting partners for each of the three ‘brothers’; Mira and Sam are the intellectuals who sometimes doubt what they’re doing and think that they don’t have to ‘kill’ EVERYTHING (Even if Mira agrees with Dean that Ruby needs to die), Dean and Lily are the loners who are nevertheless able to connect to people, and Ashley and Castiel are the ones who were ‘part’ of the supernatural long before they started hunting it (As well as Ashley’s implied status as a potential vessel).
Hope you’re going to continue this soon; the early chapters could maybe use some additional depth to explore the relationships between Dean and Lily or Sam and Mira, but, if nothing else, I am REALLY looking forward to seeing how Ashley reacts to meeting Jimmy when Castiel is temporarily ‘exorcised’ from him (Particularly since I have a theory about the impact she’d have on that particular episode’s events…).
